NanoSAM Chemistry platform presentation

The nanoSAM chemistry platform is a « grey room » localized at the Ground Floor of Building 351 door 16. It is equipped, in addition of the usual equipments of a chemistry room (wall benches, solvent cupboard, fridge-freezer…), with a controlled atmosphere glovebox, a fume cupboard, a centrifuge, a water purifier and an electrophoresis gel densitometry device. This platform is opened to external users, access should be asked to Céline Dablemont.

The nanoSAM chemistry platform has been installed during autumn 2009. It consists in a « grey room » which has been equipped, for this purpose, with an air conditioning device. It is equipped, in addition of the usual equipments of a chemistry room (wall benches, solvent cupboard, fridge-freezer…), with a controlled atmosphere glovebox, a fume cupboard allowing strong acid manipulation, a centrifuge, a water purifier which allows to obtain either demineralized water or ultra-pure water and an electrophoresis gel densitometry device. This platform is used in everyday life by members of E (Surfaces – Interfaces : Reactivity and Nanostructuration) and G (Biophysics, biophotonics, biomedical, bioenvironnement) groups. It is opened to external users as well.

The controlled atmosphere glovebox is a GP(CONCEPT)-T2 flexible model with two gloves from Jacomex which is 1.2 m width, 90 cm height and 72.5 cm depth. At the present time, it is supplied with nitrogen and purified from oxygen and water by an associated independent purification unit. Remaining oxygen and water amounts can be followed permanently thanks to oxygen and water analyzers.
This glovebox is equipped with two input / output locks : one of 60 cm length with a 40 cm diameter for input / output of bulky equipments ; the other one of 30 cm length with a 10 cm diameter for quick input / output of small equipments. Piloting of small input / output lock is manual. However, piloting of the big input / output lock is completely automated as well as the purification unit regeneration processes.
This glovebox has an activated coal filter which allows solvent use in the box.
This glovebox can be used for all manipulations of products sensitive to oxygen and / or water even dangerous because it can work in depression for experimentalist protection.

The fume cupboard which is 1.80 m width has been realized in polypropylene and its extraction circuit has been designed in order to allow manipulation, among various things, of strong acids.

The centrifuge is an Allegra X-22R model from Beckman Coulter. It allows cooling down the samples during the centrifugation and is equipped with two exchangeable rotors. The first one is a F0630 rotor which can receive until 6 tubes of 35 mL. Its maximum rotation speed is 15 300 rounds per minute, which corresponds to a 20 450 g acceleration. The second one is a F2402H rotor which can receive until 24 ependorf-type tubes of 2 mL. Its maximum rotation speed is 15 500 rounds per minute, which corresponds to a 22 065 g acceleration.

The water purifier allows obtaining either demineralized water, or ultra-pure water.
Demineralized water is obtained from city water thanks to going through a E200-type demineralization cylinder from Aquadem. This demineralized water resistivity is controlled by a mini-bip type resistivimeter from Véolia Water STI. It is higher than 5 Megaohm, which corresponds to conductivity lower than 0.2 microSiemens.
Ultra-pure water is obtained from previous demineralized water thanks to a Purelab Classic UVF polisher model from Elga / Véolia Water STI. This polisher can produce until 1.5 L.min-1 of ultra-pure water. This water resistivity is 18.2 Megaohm at 25°C ; its residual total organic carbon (TOC) is below 1 – 3 ppb ; its remaining bacteria are below 1 CFU/mL thanks to the 0.2 micron filter ; Rnase and Dnase are eliminated ; its remaining bacterial endotoxines are below 0.001 EU/mL.
